if you switch profiles in Elite 2, it will take effect immediately,
ReaSnow S1 will receive whatever the controller gives.

e.g. if you change the curve of Elite 2's sticks, it will work even if Elite 2 is connected to S1.

I tried right now to mapped Y to b and with the keyboard I pressed Y nothing happens

sorry, these did not help us to understand your current issue with Elite 2.
please provide the following details:
1. a screenshot of your settings for Elite 2 controller on XBox condole or on PC
2. the config you are using
3. how these peripherals are connected to ReaSnow S1
4. ReaSnow S1 firmware version/ APP version
5. what can you see from the HOME page when I/O monitor is enabled
